In this study, the effect of a polycaprolactone (PCL)/gelatin (GE) scaffold on differentiation of rat mesenchymal stem cells (MSCs) into hepatocyte-like cells was investigated. The characteristics of the scaffolds were analyzed by scanning electron microscopy, X-ray diffraction, and fourier transform infrared spectroscopy. The effects of the PCL/GE scaffold on cell growth, attachment, and adhesion were determined by cell counting kit-8 and crystal violet assay. Hepatocyte-like cell differentiation was evaluated by the activity of alkaline phosphatase, the expression of liver-related genes, and cell morphology and protein expression. Scanning electron microscopy showed that the PCL/GE scaffold had good uniformity and interconnected structure. X-ray diffraction and infrared spectroscopy indicated that the PCL/GE scaffold had porous structure. The PCL/GE scaffold enhanced cell growth, attachment, and adhesion of MSCs. Compared to the control group, the PCL/GE scaffold increased the expression of hepatic genes and promoted the synthesis of cell membrane proteins, as determined by the activity of alkaline phosphatase.
