Microparticles (MP) are small membrane vesicles shed from activated or apoptotic cells. Cell-derived MP have been implicated in the communication between endothelial cells and circulating stem cells, but their significance in the cellular communication between progenitor cells and endothelium in the haematopoietic system has not been studied to date. In the present study, we analysed the number of endothelial cell-derived MP (CD31+, CD146+, CD105+, CD144+) and their possible relation to CD34+ stem cells. Human umbilical vein endothelial cells (HUVEC) and blood mononuclear cells (MNC) were isolated from umbilical cord blood (UCB) and from peripheral blood (PB) of 18 healthy controls, 26 patients with acute myeloid leukaemia (AML), 21 with acute lymphoblastic leukaemia (ALL) and 23 with non-Hodgkin lymphoma (NHL). A four-colour flow cytometry was used to analyse the phenotype of the MP as well as the frequencies of the CD34+ stem cells and circulating endothelial cells. The mean number of MP/10(6) HUVEC in UCB of healthy controls was 12.9 ± 6.8 and decreased to 6.7 ± 1.6 in AML (P = 0.004), 6.9 ± 1.9 in ALL (P = 0.002) and 8.3 ± 3.3 in NHL (P = 0.074). An inverse correlation was observed between the mean number of MP and the number of CD34+ stem cells in PB of patients with haematological malignancies, in contrast to healthy controls. Our data demonstrate for the first time that in haematological malignancies MP are down-regulated in PB. Moreover, our results reveal a negative correlation between the number of MP and the frequency of CD34+ stem cells. We suggest that the number of MP is a measure
